Odd. I cant see my NPCs, they dont help me in combat, they dont block doors, yet they restrict my access to various(SP?) locates: eg. Vault city (one is a ghoul, aparently), The malamuke saloon in redding, etc, etc...
WTF?! any cures, other than start over?

You might try reinstalling and see if your savegames still work. That *might* fix it.
that happens when you sleep with one of the bishop women. happened to me...
but by that time they were just plain annoying so it was a good thing they dissaperated.
if you can spare an older save than go back to it, and tell them all to wait outside when you sleep with one of the bishops/go upstairs. i think it has to do something with how when you get called upstairs into bishops "lounge" or whatever, none of your npc's follow you. and then theres probubly a bug that stops them from coming back...
